The performance of a column depends on factors like high efficiency, good selectivity, and fast analysis speed. Commercially available columns often use different types of particulate fillers, including porous silica gel, silica-based bonded phases, alumina, organic polymer microspheres, ion exchange resins, and even porous carbon. These materials usually have particle sizes ranging from 3 to 10 micrometers, allowing for theoretical efficiencies of up to 50,000 to 160,000 per meter. For most routine analyses, a column efficiency of around 5,000 is sufficient, while more complex separations may require up to 20,000 plates. This means that typical column lengths range between 10 to 30 cm, making them suitable for analyzing complex mixtures.

A standard column consists of several key parts: the column tube, pressure cap, ferrule (or seal ring), sieve plate (filter), joint, and screw. Most column tubes are made of stainless steel, but when the operating pressure is below 70 kg/cm², thick-walled glass or quartz tubes can also be used. The inner walls must be highly smooth to minimize wall effects and improve efficiency. Some stainless steel columns are polished, while others are coated with fluoroplastic to achieve similar results. Thin-walled columns might use fused silica or glass linings. At both ends of the column, sieve plates—often made of sintered stainless steel or titanium alloy—are installed to prevent the packing material from escaping. These plates have pore sizes ranging from 0.2 to 20 micrometers, depending on the particle size of the filler used.

Columns can be classified into analytical and preparative types based on their application. Analytical columns are smaller and used for identification and quantification, while preparative columns are larger and designed for bulk sample processing. Common dimensions include conventional analytical columns (inner diameter 2–5 mm, length 10–30 cm), narrow-bore columns (inner diameter 1–2 mm, length 10–20 cm), capillary columns (inner diameter 0.2–0.5 mm), semi-preparative columns (inner diameter >5 mm), and laboratory or production preparation columns with much larger diameters. The inner diameter and length of a column are determined by the flow rate, particle size of the packing, and the need to avoid wall effects.
